Backbone hydrolysis comes up often in conversation and rarely with the context attached. Here we lay out the basics in order, then work through the practical considerations.

Identity and purity are checked by reversed-phase high-performance liquid chromatography, usually with ultraviolet detection near 214 nanometres, where the peptide bond absorbs. Mass spectrometry confirms the expected mass and reveals whether truncation products or adducts are present. Acid hydrolysis followed by amino acid analysis gives the residue ratio, which should approximate one alanine, one glutamate, one aspartate and one glycine. Counter-ions such as acetate or trifluoroacetate remain in the dried product and lower net peptide content, so a stated purity figure on a label does not by itself describe how much peptide a vial holds.
Regulatory treatment differs by country. No formulation of epitalon holds a marketing authorisation as a medicine in the United States or the European Union, where material sold for laboratory use is handled as a research chemical and is not intended for human consumption. In Russia, several short peptide preparations from the same institute's peptide series are registered medicinal products, and epitalon appears in that national context. Elsewhere it is frequently offered as a cosmetic ingredient, a category with lighter requirements. Advertising claims about longevity or disease prevention are restricted in most jurisdictions, which limits how sellers describe the compound.
Typical storage for the lyophilized powder is −20 °C or lower, in a sealed container protected from light and moisture. Hygroscopic material should be allowed to equilibrate to room temperature before the vial is opened, which limits condensation on the contents. Working solutions are commonly divided into single-use aliquots and frozen to avoid repeated freeze-thaw cycles. Dilute solutions are more prone to adsorption onto plastic surfaces and to loss during filtration, so procedures that minimize transfers and use low-binding labware are preferable.
Reversed-phase high-performance liquid chromatography is the standard approach for assessing purity, usually with ultraviolet detection near 214 nm, where the peptide bond absorbs. Mass spectrometry, most often with electrospray ionization, confirms the molecular mass and helps reveal truncation or deletion byproducts. Amino acid analysis can verify composition, and counterion content is sometimes measured because peptides purified with trifluoroacetic acid retain variable amounts of that salt. Purity figures reported without a stated method and detection wavelength are difficult to interpret.
Material sold for research use varies widely in documented quality. A useful verification package includes a certificate of analysis that states peptide content rather than only net weight, the chromatographic method and column used, and a mass spectrum consistent with the expected mass. Independent testing by a third-party laboratory is occasionally reported. Statements of identity resting only on a supplier label provide little assurance, and the gap between nominal mass and actual peptide content can be substantial once counterions and residual water are counted.

Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography with ultraviolet detection, frequently near 214 nanometers where the peptide bond absorbs. Mass spectrometry, most often with electrospray ionization, confirms the observed molecular mass and helps separate the intact peptide from truncated or modified forms. Amino acid analysis or tandem mass spectrometry can establish residue order. Purity is commonly quoted as an area percentage, yet such values are method-dependent, and comparisons between laboratories require matching column, gradient and detection wavelength.
Laboratory handling begins with dissolution of the lyophilized powder in water or a suitable aqueous buffer. The dry solid is the more stable form, so stock solutions are generally prepared only when required and kept cold afterwards. Repeated freezing and thawing of a solution is avoided because it encourages aggregation and gradual loss of the intact chain. Diluents and containers are selected to limit adsorption of a short peptide onto plastic surfaces and to reduce microbial growth in aqueous preparations.
Soy sauce (醬油, jiàngyóu) is considered almost as old as soy paste—a type of fermented paste (jiàng, 醬) obtained from soybeans—which had appeared during the Western Han dynasty (206 BC – 220 AD) and was listed in the bamboo slips found in the archaeological site Mawangdui (馬王堆). There are several precursors of soy sauce that are associated products with soy paste. The oldest is qingjiang (清醬), which appeared in AD 40 and was listed in Simin Yueling (四民月令). Others are jiangqing (醬清), chizhi (豉汁) and chiqing (豉清), which were recorded in the Qimin Yaoshu (齊民要術) in AD 540. By the time of the Song dynasty (960–1279 AD), the term jiàng yóu (醬油) had become the accepted name for the liquid condiment, documented in two books: Shanjia Qinggong (山家清供) and Pujiang Wushi Zhongkuilu (浦江吳氏中饋錄) during the Song dynasty (960–1279 AD). Like many salty condiments, soy sauce was originally a way to stretch salt, historically an expensive commodity. During the Zhou dynasty of ancient China, fermented fish with salt was used as a condiment in which soybeans were included during the fermentation process. By the time of the Han dynasty, this had been replaced with the recipe for soy paste and its by-product soy sauce, by using soybeans as the principal ingredient, with fermented fish-based sauces developing separately into fish sauce.
In the 1950s, the French established secret commando groups based on loyal Montagnard ethnic minorities referred to as "partisans" or "maquisards", called the Groupement de Commandos Mixtes Aéroportés (Composite Airborne Commando Group or GCMA), later renamed Groupement Mixte d'Intervention (GMI, or Mixed Intervention Group), directed by the SDECE counter-intelligence service. The SDECE's "Service Action" GCMA used both commando and guerrilla techniques and operated in intelligence and secret missions from 1950 to 1955. Declassified information about the GCMA includes the name of its commander, famous Colonel Roger Trinquier, and a mission on April 30, 1954, when Jedburgh veteran Captain Sassi led the Meo partisans of the GCMA Malo-Servan in Operation Condor during the siege of Dien Bien Phu. In 1951, Adjutant-Chief Vandenberghe from the 6th Colonial Infantry Regiment (6e RIC) created the "Commando Vanden" (aka "Black Tigers", aka "North Vietnam Commando #24") based in Nam Định. Recruits were volunteers from the Thổ people, Nùng people and Miao people. This commando unit wore Việt Minh black uniforms to confuse the enemy and used techniques of the experienced Bo doi (Bộ đội, regular army) and Du Kich (guerrilla unit). Việt Minh prisoners were recruited in POW camps. The commando was awarded the Croix de Guerre des TOE with palm in July 1951; however, Vandenberghe was betrayed by a Việt Minh recruit, commander Nguien Tinh Khoi (308th Division's 56th Regiment), who assassinated him (and his Vietnamese fiancée) with external help on the night of January 5, 1952.
=== Temperature === Two issues are involved when considering the temperature of a ligation reaction. First, the optimum temperature for DNA ligase activity which is 37°C, and second, the melting temperature (Tm) of the DNA ends to be ligated. The melting temperature is dependent on length and base composition of the DNA overhang—the greater the number of G and C, the higher the Tm since there are three hydrogen bonds formed between G-C base pair compared to two for A-T base pair—with some contribution from the stacking of the bases between fragments. For the ligation reaction to proceed efficiently, the ends should be stably annealed, and in ligation experiments, the Tm of the DNA ends is generally much lower than 37°C. The optimal temperature for ligating cohesive ends is therefore a compromise between the best temperature for DNA ligase activity and the Tm where the ends can associate. However, different restriction enzymes generates different ends, and the base composition of the ends produced by these enzymes may also differ, the melting temperature and therefore the optimal temperature can vary widely depending on the restriction enzymes used, and the optimum temperature for ligation may be between 4-15°C depending on the ends. Ligations also often involve ligating ends generated from different restriction enzymes in the same reaction mixture, therefore it may not be practical to select optimal temperature for a particular ligation reaction and most protocols simply choose 12-16°C, room temperature, or 4°C.
Extracellular polymeric substances (EPS) are natural polymers of high molecular weight secreted by microorganisms into their environment. EPS establish the functional and structural integrity of biofilms, and are considered the fundamental component that determines the physicochemical properties of a biofilm. EPS in the matrix of biofilms provides compositional support and protection of microbial communities from the harsh environments. Components of EPS can be of different classes of polysaccharides, lipids, nucleic acids, proteins, lipopolysaccharides, and minerals.
=== Ivermectin === Oral ivermectin is effective in eradicating scabies, often in a single dose. It is the treatment of choice for crusted scabies, and is sometimes prescribed in combination with a topical agent. It has not been tested on infants, and is not recommended for children under six years of age. Topical ivermectin preparations are effective for scabies in adults. It has also been useful for sarcoptic mange, the veterinary analog of human scabies. One review found that the efficacy of permethrin is similar to that of systemic or topical ivermectin. A separate review found that although oral ivermectin is usually effective for the treatment of scabies, it does have a higher treatment failure rate than topical permethrin. Another review found that oral ivermectin provided a reasonable balance between efficacy and safety. A study has demonstrated that scabies is markedly reduced in populations taking ivermectin regularly; the drug is widely used for treating scabies and other parasitic diseases, particularly among the poor and disadvantaged in the tropics, beginning with the developer Merck providing the drug at no cost to treat onchocerciasis from 1987.

On the "left side" of the genome there are two promoters called p5 and p19, from which two overlapping messenger ribonucleic acids (mRNAs) of different length can be produced. Each of these contains an intron which can be either spliced out or not. Given these possibilities, four various mRNAs, and consequently four various Rep proteins with overlapping sequence can be synthesized. Their names depict their sizes in kilodaltons (kDa): Rep78, Rep68, Rep52 and Rep40. Rep78 and 68 can specifically bind the hairpin formed by the ITR in the self-priming act and cleave at a specific region, designated terminal resolution site, within the hairpin. They were also shown to be necessary for the AAVS1-specific integration of the AAV genome. All four Rep proteins were shown to bind ATP and to possess helicase activity. It was also shown that they upregulate the transcription from the p40 promoter (mentioned below), but downregulate both p5 and p19 promoters.
Modified bases occur in DNA. The first of these recognized was 5-methylcytosine, which was found in the genome of Mycobacterium tuberculosis in 1925. The reason for the presence of these noncanonical bases in bacterial viruses (bacteriophages) is to avoid the restriction enzymes present in bacteria. This enzyme system acts at least in part as a molecular immune system protecting bacteria from infection by viruses. Modifications of the bases cytosine and adenine, the more common and modified DNA bases, play vital roles in the epigenetic control of gene expression in plants and animals. A number of noncanonical bases are known to occur in DNA. Most of these are modifications of the canonical bases plus uracil.
The CAMP test (Christie–Atkins–Munch-Petersen) is a test to identify group B β-hemolytic streptococci (Streptococcus agalactiae) based on their formation of a substance, CAMP factor, that enlarges the area of hemolysis formed by the β-hemolysin elaborated from Staphylococcus aureus. Although the test is usually used to identify group B streptococcus, there is some evidence that the CAMP factor gene is present in several groups of streptococci, including group A. CAMP factor is a phospholipase produced by Group B Streptococci (Streptococcus agalactiae) that enhances hemolysis of Staphylococcus aureus. A similar factor has been identified in Bartonella henselae. The CAMP test can be used to identify Streptococcus agalactiae. Though not strongly beta-hemolytic on its own, group B strep presents with wedge-shaped colonies in the presence of Staphylococcus aureus. It can also be used to identify Listeria monocytogenes which produces a positive CAMP reaction.

In July 2008, the whole site was rebranded under the Edge title, as that was the senior of the two brands. In May 2014 it was reported that Future intended to close the websites of Edge, Computer and Video Games and their other videogame publications; in December 2014, it was confirmed that the C&VG website would close and its content would instead be published at GamesRadar, and in January 2015, it was announced that the same would happen to the Edge website. Between 2015 and 2018, Edge articles were occasionally republished on Kotaku UK. Edge has been redesigned three times since the magazine launched. The first redesign occurred in 1999; the second in 2004; and the third in 2011. The first redesign altered the magazine's dimensions to be wider than the original shape. The latest design changes the magazine's physical dimensions for the second time, and introduces a higher quality of paper stock than was previously used.

"have a sensitivity of at least 80% for detecting a particular psychiatric disorder" "should have a specificity of at least 80% for distinguishing this disorder from other psychiatric or medical disorders" "should be reliable, reproducible, and ideally be noninvasive, simple to perform, and inexpensive" "proposed biomarkers should be verified by 2 independent studies each by a different investigator and different population samples and published in a peer-reviewed journal" The review concluded that although neuroimaging diagnosis may technically be feasible, very large studies are needed to evaluate specific biomarkers which were not available.
=== Healthcare === In order to bridge the gaps in Indian healthcare, Ambani launched the Kokilaben Dhirubhai Ambani Hospital & Medical Research Institute (KDAH) in Mumbai in 2009, a quaternary care facility. It is the only hospital in Mumbai to receive accreditation from the JCI (Joint Commission International, USA), NABH (National Accreditation Board for Healthcare, India), CAP (College of American Pathologists, USA) and NABL (National Accreditation Board for Laboratories, India). Other standouts include the first comprehensive centre for liver transplant and the first integrated centre for children's cardiac care in western India; its robotic surgery programme; the centres for rehabilitation and sports medicine; and its initiative to open 18 cancer care centres in rural Maharashtra.
Throughout human history, intentional application of poison has been used as a method of murder, pest-control, suicide, and execution. As a method of execution, poison has been ingested, as the ancient Athenians did (see Socrates), inhaled, as with carbon monoxide or hydrogen cyanide (see gas chamber), injected (see lethal injection), or even as an enema. Poison's lethal effect can be combined with its allegedly magical powers; an example is the Chinese gu poison. Poison was also employed in gunpowder warfare. For example, the 14th-century Chinese text of the Huolongjing written by Jiao Yu outlined the use of a poisonous gunpowder mixture to fill cast iron grenade bombs. While arsenic is a naturally occurring environmental poison, its artificial concentrate was once nicknamed inheritance powder. In Medieval Europe, it was common for monarchs to employ personal food tasters to thwart royal assassination, in the dawning age of the Apothecary.
=== Taxi === Commercial taxi services and route taxis are widely used in Moscow. During the mid-2010s, service platforms such as Yandex Taxi, Uber, and Gett displaced many private drivers and small service providers; in 2015, these platforms serviced more than 50% of all taxi orders in the city. As of 2021, Russian technology company Yandex was testing self-driving taxis in Moscow.

The solid is kept cold, dry and dark, typically at minus twenty degrees Celsius, in a sealed container with desiccant. Dissolved material is kept refrigerated and used promptly. Freezing and thawing a solution repeatedly is avoided because it accelerates degradation.
Reversed-phase chromatography with ultraviolet detection gives a purity percentage, while mass spectrometry confirms identity. Amino acid analysis checks the residue composition. Buyers typically request a certificate of analysis showing these measurements for the specific lot.
It is not authorised as a medicine in the United States or the European Union. Material is sold there as a research chemical or cosmetic ingredient. Some related short peptides are registered drugs in Russia, which is a distinct national situation rather than a general approval.
The powder is normally kept at −20 °C or below in a sealed, moisture-protected container. Letting the vial reach room temperature before opening helps prevent condensation on the contents.
